چکیده

Ovarian cancer is one of the most deadly malignancies in women because of its poor prognosis and that a majority of patients are diagnosed at advanced stage. Therefore, chemotherapy becomes the most important treatment option in most ovarian cancer cases. However, chemoresistance in relapsed cases is the major obstacle for the clinical management of this disease. Mounting evidences have suggested the de novo (intrinsic) and acquired (extrinsic) chemoresistance are two major underlying mechanisms occurring in human cancers. The de novo chemoresistance is attributed to the existence of cancer stem cells, while the genetic and/or epigenetic alterations in dysregulation of oncogenes or tumor suppressor genes contribute to the acquired chemoresistance. In this review, we will summarize and discuss the recent findings of the above mechanisms in chemoresistance and particularly, we will focus on the significance of putative miRNAs expressions and their associated signaling regulations in the development of acquired chemoresistance in ovarian cancer. *Corresponding author: David W Chan, Department of Obstetrics and Gynaecology, L747 Laboratory Block, LKS Faculty of Medicine, 21 Sassoon Road, Pokfulam, Hong Kong, Tel: (852) 2819-9367; Fax: (852) 2816-1947; E-mail: [email protected] Received May 21, 2012; Accepted May 21, 2012; Published May 24, 2012 Citation: Liu MX, Chan DW, Ngan HYS (2012) Mechanisms of Chemoresistance in Human Ovarian Cancer at a Glance. EGFR Blockade Reverses Cisplatin Resistance in Human Epithelial Ovarian Cancer Cells

Background: Epithelial ovarian cancer (EOC) is one of the most lethal gynecological malignancy worldwide. Although the majority of EOC patients achieve clinical remission after induction therapy, over 80% relapse and succumb to the chemoresistant disease. Previous investigations have demonstrated the association of epidermal growth factor receptor (EGFR) with resistance to cytotoxic chemotherap...

Generation of Cisplatin-Resistant Ovarian Cancer Cell Lines

Ovarian cancer is the most lethal gynecological cancer in which cisplatin-based treatment plays fundamental role as the first line chemotherapy option. However, development of platinum-resistance is a critical and poorly understood problem in ovarian cancer treatment. Although in vitro generation of platinum-resistant ovarian cancer cell lines is a long established approach to uncover the molec...

Human Cancer Modeling: Recapitulating Tumor Heterogeneity Towards Personalized Medicine

Despite diagnostic, preventive and therapeutic advances, growing incidence of cancer and high rate of mortality among patients affected by specific cancer types indicate current clinical measures are not ideally useful in eradicating cancer. Chemoresistance and subsequent disease relapse are believed to be mainly driven by the cell-molecular heterogeneity of human tumors that necessitates perso...

Up-Regulation of Periostin and Reactive Stroma is Associated with Primary Chemoresistance and Predicts Clinical Outcomes in Epithelial Ovarian Cancer Running title: Reactive stroma signature predicts chemoresistance

Purpose: Up to one third of ovarian cancer patients are intrinsically resistant to platinum-based treatment. However, predictive and therapeutic strategies are lacking due to a poor understanding of the underlying molecular mechanisms. This study aimed to identify key molecular characteristics that are associated with primary chemoresistance in epithelial ovarian cancers. Experimental Design: G...
